    Vibrationally excited populations from IR‐multiphoton absorption. I. Absorbed energy and reaction yield measurements

    The molecule 1,1,2‐trifluroethane (TFE) was used in experiments to determine the population distribution of excited molecules produced by infrared multiphoton absorption induced by high power TEA CO2 lasers operating at 1079.85 cm−1 [9.6 μm R(22) line]. Optoacoustic measurements of absorbed laser power provided a measure of the mean energy of the population distribution, while very low pressure photolysis measurements of the collision‐free decomposition yield gave information about the high‐energy tail of the distribution. The experimental results were accurately simulated using a Master Equation model that incorporated Quack’s statistical–dynamical theory of infrared multiphoton absorption (cases B and C), RRKM unimolecular reactions (three channels), and collisional energy transfer. The computer simulations included known TFE molecular properties and only four adjustable parameters, which were very highly constrained in order to fit the experimental data. From the simulations, we conclude that the optical coupling matrix elements are dramatically reduced in magnitude for energies above the reaction thresholds. This effect is symptomatic of the vibrational anharmonicity due to the presence of the reaction channels, even in molecules that have not yet reacted, resulting in vibrational frequency shifts of the absorption lines out of resonance with the laser line.     Inverted Fluorescence Microscope

    The Inverted Fluorescence Microscope senior project team at Cal Poly, San Luis Obispo designed, assembled, and tested a proof-of-concept inverted fluorescence microscope for the university’s Microfabrication Laboratory. Administrators of the laboratory wished to use fluorescence for research and experiments involving cell growth and flow visualization on the micro-scale, and did not have the budget to purchase one of the costly commercially available options. The scope of this design challenge was to produce a low-cost inverted fluorescence microscope employing available optical components and additional readily sourced parts to expand the use of fluorescence microscopy accessible to undergraduate students in the Microfabrication Laboratory. This document is an account of the final microscope design as well as the engineering design process, project management procedures, and timeline followed to produce a working design verification prototype. The final product successfully resolved images of microfluidic devices in brightfield mode with automated maneuverability in the X-Y plane. It is equipped with fluorescence capabilities, and will serve as a valuable, low-cost research tool and platform for future student projects

    Avian influenza viruses (AIVs) have been pivotal to the origination of human pandemic strains. Despite their scientific and public health significance, however, there remains much to be understood about the ecology and evolution of AIVs in wild birds, where major pools of genetic diversity are generated and maintained. Here, we present comparative phylodynamic analyses of human and AIVs in North America, demonstrating (i) significantly higher standing genetic diversity and (ii) phylogenetic trees with a weaker signature of immune escape in AIVs than in human viruses. To explain these differences, we performed statistical analyses to quantify the relative contribution of several potential explanations. We found that HA genetic diversity in avian viruses is determined by a combination of factors, predominantly subtype-specific differences in host immune selective pressure and the ecology of transmission (in particular, the durability of subtypes in aquatic environments). Extending this analysis using a computational model demonstrated that virus durability may lead to long-term, indirect chains of transmission that, when coupled with a short host lifespan, can generate and maintain the observed high levels of genetic diversity. Further evidence in support of this novel finding was found by demonstrating an association between subtype-specific environmental durability and predicted phylogenetic signatures: genetic diversity, variation in phylogenetic tree branch lengths, and tree height. The conclusion that environmental transmission plays an important role in the evolutionary biology of avian influenza viruses—a manifestation of the “storage effect”—highlights the potentially unpredictable impact of wildlife reservoirs for future human pandemics and the need for improved understanding of the natural ecology of these viruses

    Temporal trends in 6-minute walking distance for older Japanese adults between 1998 and 2017

    Background The 6-minute walking distance (6MWD) is an excellent measure of both functional endurance and health. The primary aim of this study was to estimate temporal trends in 6MWD for older Japanese adults between 1998 and 2017; the secondary aim was to estimate concurrent trends in body size (i.e., height and mass) and self-reported participation in exercise/sport. Methods Adults aged 65–79 years were included. Annual nationally representative 6MWD data (n = 103,505) for the entire period were obtained from the Japanese Ministry of Education, Culture, Sports, Science and Technology. Temporal trends in means (and relative frequencies) were estimated at the gender–age level by best-fitting sample-weighted linear/polynomial regression models, with national trends estimated by a post-stratified population-weighting procedure. Temporal trends in distributional variability were estimated as the ratio of coefficients of variation. Results Between 1998 and 2017 there was a steady, moderate improvement in mean 6MWD (absolute = 45 m (95% confidence interval (CI): 43–47); percent = 8.0% (95%CI: 7.6–8.4); effect size = 0.51 (95%CI: 0.48–0.54)). Gender- and age-related temporal differences in means were negligible. Variability in 6MWD declined substantially (ratio of coefficients of variation = 0.89, 95%CI: 0.87–0.92), with declines larger for women compared to men, and for 75–79-year-olds compared to 65–74-year-olds. Correspondingly, there were moderate and negligible increases in mean height and mass, respectively, and negligible increases in the percentage who participated in exercise/sport at least 3 days per week and at least 30 min per session. Conclusions There has been a steady, moderate improvement in mean 6MWD for older Japanese adults since 1998, which is suggestive of corresponding improvements in both functional endurance and health. The substantial decline in variability indicates that the temporal improvement in mean 6MWD was not uniform across the distribution. Trends in 6MWD are probably influenced by corresponding trends in body size and/or participation in exercise/sport

    Commercialising the production of Cobia in Australia

    This project is a collaboration between the Department of Agriculture and Fisheries (DAF) and the Cobia aquaculture sector, predominantly Pacific Reef Fisheries (PRF). It was undertaken to consolidate the aquaculture in Australia of Cobia, a species offering considerable potential as a diversification option for pond-based culture activities in Queensland. The project addressed key elements of the production cycle in order to move industry towards a more commercial footing and advance both the scientific knowledge and human capabilities of the sector. The project involved DAF staff at the Department’s Bribie Island Research Centre (BIRC) and staff from the PRF farm in Ayr, North Queensland. From 2014-2017, research examined: health issues affecting Cobia in culture both in the hatchery and on-farm; the emerging issue of intersex in Cobia and potential for this to further impact production; capability development of PRF staff in preparation of the establishment of a commercial Cobia hatchery; and, the involvement of some new entrants to Cobia aquaculture utilising tank-based production systems

    Restenosis and risk of stroke after stenting or endarterectomy for symptomatic carotid stenosis in the International Carotid Stenting Study (ICSS): secondary analysis of a randomised trial.

    BACKGROUND: The risk of stroke associated with carotid artery restenosis after stenting or endarterectomy is unclear. We aimed to compare the long-term risk of restenosis after these treatments and to investigate if restenosis causes stroke in a secondary analysis of the International Carotid Stenting Study (ICSS). METHODS: ICSS is a parallel-group randomised trial at 50 tertiary care centres in Europe, Australia, New Zealand, and Canada. Patients aged 40 years or older with symptomatic carotid stenosis measuring 50% or more were randomly assigned either stenting or endarterectomy in a 1:1 ratio. Randomisation was computer-generated and done centrally, with allocation by telephone or fax, stratified by centre, and with minimisation for sex, age, side of stenosis, and occlusion of the contralateral carotid artery. Patients were followed up both clinically and with carotid duplex ultrasound at baseline, 30 days after treatment, 6 months after randomisation, then annually for up to 10 years. We included patients whose assigned treatment was completed and who had at least one ultrasound examination after treatment. Restenosis was defined as any narrowing of the treated artery measuring 50% or more (at least moderate) or 70% or more (severe), or occlusion of the artery. The degree of restenosis based on ultrasound velocities and clinical outcome events were adjudicated centrally; assessors were masked to treatment assignment. Restenosis was analysed using interval-censored models and its association with later ipsilateral stroke using Cox regression. This trial is registered with the ISRCTN registry, number ISRCTN25337470. This report presents a secondary analysis, and follow-up is complete. FINDINGS: Between May, 2001, and October, 2008, 1713 patients were enrolled and randomly allocated treatment (855 were assigned stenting and 858 endarterectomy), of whom 1530 individuals were followed up with ultrasound (737 assigned stenting and 793 endarterectomy) for a median of 4·0 years (IQR 2·3-5·0). At least moderate restenosis (≥50%) occurred in 274 patients after stenting (cumulative 5-year risk 40·7%) and in 217 after endarterectomy (29·6%; unadjusted hazard ratio [HR] 1·43, 95% CI 1·21-1·72; p<0·0001). Patients with at least moderate restenosis (≥50%) had a higher risk of ipsilateral stroke than did individuals without restenosis in the overall patient population (HR 3·18, 95% CI 1·52-6·67; p=0·002) and in the endarterectomy group alone (5·75, 1·80-18·33; p=0·003), but no significant increase in stroke risk after restenosis was recorded in the stenting group (2·03, 0·77-5·37; p=0·154; p=0·10 for interaction with treatment). No difference was noted in the risk of severe restenosis (≥70%) or subsequent stroke between the two treatment groups. INTERPRETATION: At least moderate (≥50%) restenosis occurred more frequently after stenting than after endarterectomy and increased the risk for ipsilateral stroke in the overall population. Whether the restenosis-mediated risk of stroke differs between stenting and endarterectomy requires further research.     Work-Unit Absenteeism: Effects of Satisfaction, Commitment, Labor Market Conditions, and Time

    Prior research is limited in explaining absenteeism at the unit level and over time. We developed and tested a model of unit-level absenteeism using five waves of data collected over six years from 115 work units in a large state agency. Unit-level job satisfaction, organizational commitment, and local unemployment were modeled as time-varying predictors of absenteeism. Shared satisfaction and commitment interacted in predicting absenteeism but were not related to the rate of change in absenteeism over time. Unit-level satisfaction and commitment were more strongly related to absenteeism when units were located in areas with plentiful job alternatives
